360名中国汉族PD患者(63.5±10.4年)被纳入队列。所有患者均来自中国呼和浩特内蒙古医科大学附属医院和中国巴彦淖尔市医院,2015年至2018年。所有患者生活在中国西北地区,彼此之间没有亲属关系。所有患者均由经验丰富的神经科医生检查,PD诊断依据临床标准[

采用血液DNA提取试剂盒(TIANamp blood DNA kit;天根生物技术,北京,中国),储存在−20°C。采用聚合酶链反应(PCR)/连接酶检测反应法进行基因分型。引物由上海海宇生物工程有限公司合成;引物信息如表所示

在此,我们对752名西北汉族人群进行病例对照研究,进一步探讨SLC6A3 3 '末端在PD发生中的作用。我们发现只有SLC6A3变异rs40184在基因型和等位基因上有统计学差异,这可能与PD有关。在西北汉族人群中,rs40184等位基因A可能导致PD发病风险增加,可能是PD的一个标志物。其余16个多态性位点未发现与PD相关。其他16个位点的阴性结果可能与本研究样本数量不足有关,也可能是汉族人群中罕见的基因型或等位基因。在相关研究中,单倍型分析被认为比单核苷酸多态性位点更有力[
目前,已知有1500多个基因与疾病的发生密切相关。长期以来,编码区一直是遗传疾病的主要研究方向,只有一小部分非编码区被证实是帮助基因开启和关闭以调控基因表达的有用成分。基因的正常表达离不开调控元件的参与。某些调控成分的异常也会导致相应基因编码区域的突变。因此,我们可以假设在我们的研究中,SLC6A3非编码区的变异rs40184可能导致PD易感性的差异。
总之,我们的研究结果表明,西北汉族人群中SLC6A3基因变异体rs40184的3′端与PD之间存在联系。鉴于不同人群的遗传异质性和突变具有特定的群体频率,需要进行更大的样本量研究,以确认SLC6A3变异体3′端与PD之间的相关性此外,需要对SLC6A3的3′端进行功能研究,以进一步了解SLC6A3在PD中的作用。进一步的研究可能会发现对PD具有风险或保护作用的遗传变异,这对PD的预防和治疗非常重要。
